During the past decade, researchers have reported the culture of Leydig cells and Sertoli cells in serum-free medium that contained insulin, transferrin, epidermal growth factor (EGF), leutinizing hormone or follicle stimulating hormone, somatomedin and growth hormone. A 1:1 mixture of Dulbecco’s Modified Eagle’s Medium (DMEM) and Ham’s F-12 Nutrient Mixture is found to be most satisfactory for studies of this type.\u003c\/p\u003e\n\u003cp\u003e\u003cstrong\u003eLM 002-01 \u003c\/strong\u003eand \u003cstrong\u003eLM 002-02\u003c\/strong\u003e contain, instead of serum, a supplement of nutrients, growth factors and hormones, but no L-glutamine. HEPES buffer is included in the formulation at a final concentration of 15 mM to compensate for the loss of buffering capacity incurred by eliminating serum. If necessary, add aseptically 15 mL of sterilie 200 mL L-glutamine solution (\u003cstrong\u003eLS 002-01\u003c\/strong\u003e) per 1 L of medium before use (the final concentration of L-glutamine is 2.5 mM or 365 mg\/L). \u003cstrong\u003eLM 002-01\u003c\/strong\u003e contains phenol red, but \u003cstrong\u003eLM 002-02\u003c\/strong\u003e does not. The selection of a nutrient medium is strongly influenced by (1) type of cell, (2) type of culture (monolayer, suspension, or clonal) and (3) degree of chemical definition necessary.
